چکیده

Introduction The automation of cell culture processes becomes more important in the pharmaceutical industry due to compressed timelines and the need to develop more products more efficiently. This drive to develop new processes faster and more efficient requires a streamlined workflow. Resource intensive approaches like the use of shake flasks limit the accessible design space for the development of highly productive processes or the characterization of established processes. Process automation provides the appropriate tools to address the following key points: Increasing experimental throughput ⇨ “Design of Experiments” using full factorial designs Increasing process information ⇨ improve process understanding (“Quality by Design”) Automate repetitive manual work ⇨ gain efficiency, focus on high value tasks Improve reproducibility ⇨ ensure robust processes A robotic plate handler based system was selected to meet the demands of a flexible, fast and modular screening system as presented in figure 1.

Development and application of an automated, multiwell plate based screening system for suspension cell culture

Introduction The already presented automated, multiwell plate (MWP) based screening system for suspension cell culture is now routinely used in process development. It is characterized by a fully automated workflow with integrated analytical instrumentation and uses shaken 6-24 well plates as bioreactors which can be run in batch and fed-batch mode with a capacity of up to 384 reactors in paral...

Cyclic olefin polymers: innovative materials for high-density multiwell plates.

Extension of ultra-high-throughput experiment (UHTE) approaches to new assay methodologies is often limited by compromised data quality when samples are miniaturized. Overcoming this challenge requires attending to all components of an automated laboratory system contributing to assay variability. A key but often neglected source is the high-density multiwell platform or microtiter plate. Mater...

Cell-based multiwell assays for the detection of substrate accumulation and oxidation.

We describe multiwell assays for detecting the accumulation as well as the subsequent oxidation of (14)C-labeled substrates in cultured cells. Accumulation is monitored in real time by an established scintillation proximity assay in which the scintillator is embedded in the plate base primarily detecting cell-associated radiolabel. The substrate oxidation assay is a novel variant of previously ...

Perfused multiwell plate for 3D liver tissue engineering.

In vitro models that capture the complexity of in vivo tissue and organ behaviors in a scalable and easy-to-use format are desirable for drug discovery. To address this, we have developed a bioreactor that fosters maintenance of 3D tissue cultures under constant perfusion and we have integrated multiple bioreactors into an array in a multiwell plate format. All bioreactors are fluidically isola...

Digital microfluidics (DMF), a fluid-handling technique in which picolitre-microlitre droplets are manipulated electrostatically on an array of electrodes, has recently become popular for applications in chemistry and biology. DMF devices are reconfigurable, have no moving parts, and are compatible with conventional high-throughput screening infrastructure (e.g., multiwell plate readers). For t...
